This Research Associate position is in the laboratory of Dr. Amanda Dudek in the Department of Microbiology and Immunology. We are seeking a motivated and curious research associate to join our lab focusing on primary immune cell models of HIV-1 infection. This is an exciting opportunity for recent graduates to work directly with the PI as the lab is becoming established and growing, with the opportunity for growth and independent research project(s) as skills are mastered.  The candidate will primarily be responsible for assisting with PBMC purification from healthy blood donors and immune cell subset solation, Cas9/AAV-based editing, long term primary cell differentiation protocols (14 to 36 days in culture depending on cell type), and various molecular biology techniques including molecular cloning, plasmid preparation, and quantification of editing outcomes.


Position Responsibilities

  • Research reagent ordering and lab organization
  • PBMC (peripheral blood mononuclear cell) isolation from healthy human blood donors
  • MACS (magnet activated cell separation) of primary immune cells from PBMCs
  • CRISPR-based gene editing of human immune cells and cell lines
  • Molecular biology methods to assess editing rates (PCR, sanger sequencing, TIDE/ICE analysis of knockout efficiency, digital droplet PCR quantification of knocking efficiency)
  • Primary cell culture (such as T cells, B cells, Natural Killer cells, Hematopoietic Stem and Progenitor Cells) and cytokine differentiation/polarization
  • FACS (fluorescence activated cell sorting) and fluorescence-based analysis
  • Data analysis and interpretation Perform library and online research on specified topics for use in experimentation
  • Present research progress at laboratory meetings
  • Assist in the preparation of manuscripts and grants by organizing data and preparing images, figures, and tables as directed
  • Assist in completion of required paperwork for new and departing laboratory personnel, including required vaccinations and exposure testing, annual training, radiation safety, biohazard safety, laboratory safety, and fire safety training
  • Maintain accurate records of laboratory databases (equipment and supplies, reagents, mammalian and microbial strains and stocks)
  • Assist in setting up a functional and organized laboratory and tissue culture space
  • Maintain laboratory supplies and equipment, including the timely ordering, receiving, and proper storage of supplies and equipment.
  • Maintain general laboratory functionality, including all equipment
  • Maintain a clean work area
  • Oversee undergraduate and graduate students in successful completion of assigned tasks and research

Type of Position: Specified Term. Initial appointment is for one year. Appointment may be extended based on performance and availability of funding.

Education Requirements 

  • Requires a bachelor’s degree in Microbiology, Immunology, Bioengineering, or a related field

Experience Requirements

  • A minimum of 1 year (part or full time) of experience in laboratory research
  • At least 6 months of experience with mammalian cell culture
  • Excellent aseptic and sterile technique
  • Experience with basic molecular biology techniques including PCR, DNA gel electrophoresis, molecular cloning, and an understanding of Cas9-based gene editing systems
  • Experience with protein gel electrophoresis, Western blotting, flow cytometry, and use of AAV and/or lentivirus based viral vectors is a plus but not a prerequisite
  • Experience gathering, recording, and analyzing experimental data
  • Experience with data analysis and presentation software, such as Microsoft Excel/Word, Prism, etc

Desired Qualifications

  • Prior experience with tissue culture
  • Prior experience working with viruses
  • Prior experience performing flow cytometry
  • Excellent organizational skills
